Deshmukh was born on 17 December 1978 in Mumbai to Vilasrao Deshmukh, the former chief minister of Maharashtra and Union Minister of Heavy Industries and Public Enterprises in India, and Vaishali Deshmukh. His elder brother, Amit Deshmukh, serves as an MLA from Latur City, while his younger brother, Dhiraj Deshmukh, is an MLA from Latur Rural. Aditi Deshmukh, his elder sister-in-law, is an actress, and Deepshikha Deshmukh, his younger sister-in-law, works as a film producer.
Deshmukh attended G. D. Somani Memorial School and obtained an architectural degree from Kamla Raheja College of Architecture in Mumbai. After spending a year practicing with an overseas architectural firm, he returned to India and continued his career in design. Deshmukh still holds ownership in Evolutions, an architectural and interior designing firm based in India.

Riteish started his acting career in 2003 with “Tujhe Meri Kasam” alongside Genelia D’Souza, who later became his wife. He found success in 2004 with movies like “Masti” and “Bardaasht.” His role in “Masti” got him nominated for awards like the Filmfare Award for Best Supporting Actor. After that, he starred in hit films like “Heyy Babyy,” “Dhamaal,” “House Full,” “Double Dhamaal,” “Tere Naal Love Ho Gaya,” “Housefull 2,” “Humshakals,” and “Ek Villian.”
In 2013, Riteish stepped into film production with the successful Marathi film “Balak Palak,” directed by Ravi Jadhav. The next year, he made his debut in Marathi cinema as an actor in the action film “Lai Bhaari.”
In 2022, Deshmukh directed his first film, a Marathi romantic drama called Ved. He starred alongside his wife, Genelia D’Souza. The movie is a remake of the 2019 Telugu film Majili, originally written and directed by Shiva Nirvana.
